Looks like there's a Greyhound pretty much straight from Denver to Laramie that takes about 2.5 hours. Probably your best bet.

Laramie to Jackson Hole is a bit trickier. There's technically a Greyhound route, but it leaves at 2:15 a.m., travels through 4 different states with two layovers, and takes 30 hours.

If you can get yourself to Salt Lake City (Greyhound goes there from Laramie without a bus change), there's a daily shuttle that will take you to Jackson Hole.

The Greyhound to SLC only leaves twice a day from Laramie, and the shuttle to Jackson Hole is only once a day, so the scheduling might get tricky, but it looks possible.
